A standard Subclass 485 visa generally cannot be indefinitely extended, though a second 485 for 1 to 2 more years is possible if you studied and worked in a designated regional area. For permanent residency, you must qualify through skilled migration, employer sponsorship, or a regional or partner visa pathway.
Once your three year post-study work visa is ending, you have two separate questions to answer: whether you can extend your current visa, and how you actually transition to permanent residency. The pathways differ quite a bit.

Why Regional Study Matters
Broad, automatic extensions for select degrees are no longer available, so if you studied in a major metropolitan centre without any regional footprint, a direct 485 extension is generally not an option for you. Regional study, however, can also help you qualify for the Subclass 491 provisional visa, which leads to the Subclass 191 permanent residence pathway after meeting your work and residence timeframe.
